In ambito comunitario, per valutare la qualità e la genuinità di succhi e puree di frutta si fa riferimento al Code of Practice (COP) del AIJN.
Linee di guida di riferimento che riportano le caratteristiche compositive di frutti processati industrialmente
Linee guida di riferimento individuali per 25 succhi/puree di frutta ed
ortaggi : Acerola, mela, albicocca, aronia, banana, ribes nero, mirtillo rosso, uva, pompelmo, guava, kiwi, limone, mandarino, mango, arancia, passion fruit, pesca, pera, ananas, melograno, lampone, amarena, fragola, pomodoro, carota.

AIJN Reference Guidelines
Sezione A : Requisiti qualitativi e igienici Sezione B : Parametri per la valutazione della genuinità (acidità totale, acidi organici, numero di formolo, ceneri, minerali, zuccheri..)
I parametri sono divisi in 4 categorie:
1. Parametri industriali: densità relativa e corrispondente valore di Brix per succhi naturali e succhi da
concentrato;
2. Requisiti igienici: acidità volatile, etanolo, acido D-L lattico e patulina;
3. Contaminanti ambientali: arsenico e metalli pesanti;
4. Parametri legati alla composizione: acidoL-ascorbico, olii essenziali, HMF
AIJN Reference Guidelines
Per ciascun parametro analitico presente nelle linee guida AIJN è
riportato un intervallo di variazione (valore minimo e massimo).
‘It is not possible to establish any universally applicable standards for processed fruit juices and for their authenticity. On the other hand, it is possible to give minimum or maximum values and/or ranges for the individual constituents. ‘Interpretation of data from profiling procedures Over the last 10 years a number of profiling or “fingerprinting” methods have been developed that are very useful in assessing the quality and authenticity of fruit juices/purées. In most of these methods the pattern of peaks is often more important than their actual size. The fingerprinting methods include oligosaccharide, anthocyanin and polyphenol profiles and the profile of volatile flavour compounds. The pattern of isotopic data in a sample can also be useful to enhance the detection of added sugars and/or acids or in the assessment of country of origin.’ Code of Practice, Specific Comments category B parameter
‘Additional analyses and/or traceability survey (identification of origin, variety and/or actual climatic conditions, etc.) must be undertaken in order to determine whether the deviations are specific to the particular raw material or are attributable to a processing or adulteration procedure.’ Code of Practice, General Comments
‘Polyphenol profiles These methods have particular use in the detection
of the presence of one type of fruit in another. They
again involve the separation of the compounds by
reverse phase HPLC.
The methods produce a pattern of peaks that can
be compared with a reference sample of fruit. These
patterns are generally very complex. However, it is
often only particular regions of the profile that are of
specific interest in detecting the addition of one fruit
to another.
Typical examples of the use of these methods are:
Differentiation of citrus juices
The detection of mixtures of citrus juices such as
sweet (C. sinensis) and sour (C. aurantium) orange
The presence of grapefruit in orange
Differentiation of lime from lemon.

Lo studio è stato condotto su :
Albicocche e pesche sono state trasformate in puree da una ditta conserviera italiana
Un campione medio costituito da circa 1 kg di purea è stato prelevato da ogni batch produttivo ed inviato alla SSICA

Preparazione del campione
Le puree sono state diluite con una soluzione idroalcolica al 50% di
metanolo, filtrate su filtro di cellulosa e su filtro siringa in nylon da 0,2 µm.
Il filtrato è stato sottoposto ad analisi UHPLC-HRMS/MS.
Analisi UHPLC-MS/MS
L’analisi è stata condotta con un UHPLC Thermo Scientific™ Dionex™ UltiMate 3000 RS costituito da: Pompe: sistema quaternario (HPG-3000RS) Autocampionatore (WPS-3000 TRS) Alloggiamento Colonna TCC-3000RS
Accoppiato con uno spettrometro di massa Thermo Scientific™ Q Exactive™ quadrupole-Orbitrap™ e sorgente H-ESI II.
L’acquisizione degli spettri e dei cromatogrammi e la gestione del sistema cromatografico sono state eseguite con il software Thermo Scientific™ Xcalibur™ 2.1

i principali polifenoli identificati nelle puree esaminate. Iperoside e
kanferolo-3-rutinoside sono presenti in piccole quantità.
Purea di pesca
Acido clorogenico, catechina, iperoside sono i principali polifenoli
identificati. La rutina è presente in piccole quantità; in tracce (<1 ppm) il
Kanferolo-3-glucoside.
L’analisi target non è sufficiente per individuare piccole aggiunte
di purea di pesca a purea di albicocca
Approccio untarget All datasets from Q Exactive analysis were processed with Thermo Scientific™
SIEVETM 2.1 software through the Chromatographic Alignment and Framing
algorithm.
SIEVE 2.1 software uses MS intensity from raw LC/MS data to find abundance
differences. It used a proprietary process to generate a unique “frame” for each
group of peaks within a specified m/z and retention time range. Frame by frame,
the algorithm determined whether there is a statistically significant difference in
abundance between samples. The chosen type of experiment was the Two Sample
Differential Analysis .
The apricot group is define as the control group and the application compares
the other group to this one. For each trend point, the application calculates a ratio
against the control group.

Approccio untarget Identify
Polyphenols putative identification was performed using two-stage approach:
1. Accurate mass determination generating elemental composition within a narrow
mass tolerance window for identification based on accurate precursor mass.
2. HCD product ion data to elaborate hypothetical fragmentation patterns. HCD
MS/MS spectra shown were also compared with data in literature and in the
Reference Library of m/z Cloud – Advanced mass spectral database (HighChem
LLC, Slovakia).
For some signals, the putative identification was confirmed by analysis performed on
authentic standard.
