Risposta T efficace • I linfociti T vergini devono entrare in contatto con l’Ag:
– APC catturano l’Ag e lo concentrano nei linfonodi periferici attraverso cui i linfociti T naive ricircolano
• Ad Ag provenienti da compartimento extracellulare o intracellulare deve rispondere la popolazione di linfociti T “giusta” ossia CD4 e CD8 rispettivamente:
– selettività del legame di CD8 e CD4 rispettivamente per MHC classe I e II e alla separazione della via di presentazione dell’antigene in classe II (Agsendosomiali) e in classe I (Ags citoplasmatici)
• I linfociti T devono interagire con le APC per un periodo sufficientemente lungo ad attivarsi:
– le molecole di adesione stabilizzano il legame tra APC e linfociti T garantiscono che questo avvenga
• Necessità che i linfociti T rispondano agli antigeni dei microbi, ma non a proteine innocue, magari autologhe:
– la selettività è garantita dal fatto che l’attivazione dei T richiede co-stimolatoriprodotti dalle APC solo a seguito del contatto con i microrganismi
• Il riconoscimento dell’Ag da parte di un numero esiguo di linfociti T deve riverberarsi in una risposta immunitaria sufficientemente ampia per garantirne l’efficacia:
– questo fenomeno può verificarsi grazie agli innumerevoli circuiti di amplificazione dell’attivazione T indotti sia dai microbi che dai linfociti T

Ruolo del CD40 nell’attivazione dei linfociti T
CD40L engages CD40 on the APC and may stimulate the expression of B7 molecules and the secretion of cytokines that activate T cells.Thus, CD40L on the T cells makes the APCs "better" APCs, and the B7 and CD40 pathways stimulate each other.
Naive T cells are activated by peptide-MHC complexes on antigen-presenting cells (APCs) previously activated by the binding of pathogen-associated molecular patterns (PAMP) to Toll-like receptors (TLRs).
Antigen recognition by T cells in conjunction with CD28 activation induces the expression of CD40 ligand (CD40L) on activated T cells.

Intracellular signaling events during T cell activation. Binding of the TCR and coreceptors to peptide-MHC complexes on the antigen-presenting cell (APC) initiates proximal signaling events, which result in phosphorylation of the ζ chain, binding and activation of ZAP-70, phosphorylation of adapter proteins, and activation of various cellular enzymes. These enzymes then activate transcription factors that stimulate the expression of various genes involved in T cell responses.

Declino delle risposte immunitarie
• Una volta eradicata l’infezione e rimosso lo stimolo antigenico principale molte cellule che avevano proliferato in risposta all’Ag vengono private dei fattori necessari alla loro crescita e muoiono per apoptosi.
• L’unica traccia che rimane della risposta immunitaria è la sopravvivenza dei linfociti T memoria
