APCs link innate and T-cell–mediated immunity . Pathogens or necrotic/damaged tissues are detected byAPCs, such as dendritic cells (DCs) and resident tissue m acrophages, through pattern recognitionreceptors (PRRs) such as toll-like and scavenger receptors. Signal transduction by way of thesereceptors leads to DC maturation and activates a wide arr ay of interconnected and synergistic hostdefense mechanisms. DC maturation involves upregulation of class I and II MHC molecules and costimulatory molecules, such as CD80 and CD86, and, t herefore, enables the DCs to present antigen toT cells . B) T cells that are stimulated through the TCR, after specifically binding to the antigenic peptide-MHC complex, and through CD28, after binding to B7 molecules (CD80, CD86) on APC, generate multiple biochemical signals thatresult in new gene transcription and augmented stability of mRNAs thatencode T-cell cytokines and cell surface molecules. T cells that receive twosignals differentiate into helper or effector T cells. EXAMPLE: epithelial salivary cells express B7 in Sjögren’s syn drome
Afferent costimulatory molecule pairs on T cells
T-cell surface receptors of the immunoglobulin superfam ily include CD28, ICOS, CTLA-4, and PD-1.CD28 and ICOS transduce positive costim. signals, while CTLA-4 and PD-1 negative signals to the T cell. CD28 is expressed constitutively, whereas ICOS, CTLA-4, and PD-1 are expressed after T-cell activation. CD28 and CTLA-4 share at least two ligands, CD80 (B7-1) and CD86 (B7-2). In the early phase of a T-cell–mediated immune response, CD28 will bind preferentially to B7-2 and mediate positive costimulatory signals. Later in a T-cell–mediated response, when CTLA-4 is expressed on the cell surface, CTLA-4 will compete withCD28 for binding B7-1 (expressed later) and B7-2, and T-cell activation will be modulated negatively.
